- W2318151567 abstract "Background: Hepatocyte transplantation using porous matrices is under investigation as alternative therapy for certain liver diseases. For this purpose, long-term function of transplanted hepatocytes is mandatory. This problem has not been sufficiently investigated yet. Materials and methods: In this study Lewis rats were used as donors and recipients. The liver damage was induced by CCL4 intraperitoneal injection. The animals of group I have received 106 hepatocytes on the biodegradable polymer scaffolds intraperitonealy. In the animals of II group the liver microfragments were placed into synthetic capsules and implanted into the peritoneal cavity. The animals of III group received empy capsules. All animals were studied for various periods. Results: Acute liver failure resulted in a 20% to 30% host survival. In groups 1 and 2 transplantation of hepatocytes on the biodegradable polymer scaffolds and liver microfragments on synthetic capsules significantly increased survival rates to 64% and 82%, respectively (P < .01). For group III survival was unmodified. Conclusion: After induction of ALF in rats, intraperitoneal transplantation of hepatocytes on the biodegradable polymer scaffolds and liver microfragments on synthetic capsules significantly improved survival. These results indicate that hepatocytes and liver microfragments obtained by mechanical dissociation of liver can be successfully encapsulated and transplanted into recipients with ALF to sustain liver metabolic functions." @default.
