- W2318546985 abstract "P046 Human T-cell leukemia virus type I (HTLV-I) is associated with a large spectrum of clinical manifestations, including adult T cell leukemia (ATLL) and tropical spastic paraparesis (TSP/HAM). HTLV-I neuropathogenesis is still incompletely understood. Development of TSP is dependent on the ability of HTLV-I to cross the blood-brain barrier. Provirus and viral mRNA were detected in the CNS of TSP patients but the exact mechanism(s) of HTLV-I entry into the CNS is not completely resolved. To elucidate this point we developed an in vitro model of the blood-brain barrier (BBB) in which brain endothelial cells were exposed to HTLV-I infected cells, mimicking the in vivo situation. The present study suggests that HTLV-I may cross endothelial barrier by several, not exclusive, mechanisms including i) direct passage of infected lymphocytes after rupture of the monolayer integrity, ii) transcytosis by endosomes-like vesicles through which HTLV-I crosses the endothelium, iii) productive infection of endothelial cells. Endothelium contact with HTLV-I infected lymphocytes results in the BBB injury, as shown by increased permeability data. Impaired endothelial cells function was linked to enhanced adhesion and transmigration of infected lymphocytes across the barrier. Contact between endothelial cells and HTLV-I infected lymphocytes results also in a massive and rapid budding of virions from infected cells followed by their internalisation into endosome-like structures, indicating that HTLV-I particles can cross brain barrier using transcytosis pathway. Our study also demonstrate that, after fusion with infected lymphocytes, brain endothelial cells develop a transient productive HTLV-I infection. The relevance of these data to the neuropathogenesis linked with HTLV infection will be discussed." @default.
