- W2320062913 abstract "Agouti-related protein (AgRP), an alpha-melanocyte stimulating hormone antagonist, is a signaling peptide that affects feeding behavior, energy homeostasis and adiposity. AgRP has also been shown to stimulate the hypothalamic-pituitary-adrenal (HPA) axis and augment the HPA axis response to interleukin-1. PURPOSE: To determine the effects of 90 min of treadmill exercise on circulating AgRP concentrations and the relationship of AgRP responses to cortisol. METHODS: Seven young males (mean ± SE age, BMI, VO2max, 22.6 ± 0.61y, 23.26 ± 1.44 kg/m2, 54.61 ± 3.57 ml/kg/min) completed a preliminary trial followed by an experimental trial and a control trial one month apart. Descriptive data were determined in a preliminary trial. An experimental trial began 2.5h after consumption of a standard nutrient beverage and consisted of treadmill exercise at 60% of previously determined VO2max for 90 minutes. Blood samples were collected beginning at 9:00am before (-30 and 0 min), during (18, 36, 54, 72, & 90 min), and following exercise (20, 40, & 60 min). For control conditions, blood samples were collected in a resting trial at the same time points as the experimental trial. The experimental and control trials were counterbalanced. Plasma was analyzed for AgRP (ELISA, R&D Systems, Minneapolis, MN) and cortisol (Immulite; Seimens, Los Angeles, CA). RESULTS: Although AgRP increased from 89.76 ± 7.61 pg/mL at 18 min of exercise to peak at 105.32 ± 17.43 pg/mL at 90 min, these increases were not significantly different than values in the control trial. Cortisol increased from 11.06± 1.46 μg/dL at 18 min to peak at 15.17 ± 2.09 μg/dL at 20 min recovery. Exercise values were significantly higher than control values. Moreover, at 54, 72 and 90 min during exercise and 20, 40 & 60 min of recovery, AgRP concentrations were positively correlated (p<0.05, range of r= 0.59 to 0.77) with cortisol concentrations at 90 min of exercise as well as 20 and 40 min of recovery. CONCLUSIONS: Data suggest that elevations in AgRP concentrations are associated with increases in cortisol during prolonged steady-state exercise in males and further study is required to determine whether there is a causal AgRP effect." @default.
