- W2320248893 abstract "アレルギー疾患は遺伝要因と環境要因とが複雑に関与して生じる炎症性疾患である。近年のHapMapプロジェクトによる情報基盤の整備，及び高速大量タイピング技術の確立により，大規模で信頼性の高いゲノムワイド症例対照関連解析（GWAS）が行なわれるようになってきた。アレルギーの分野でも遺伝要因が明らかとなってきている。その結果，疾患関連領域には，IL-33，その受容体であるIL1R1，TSLPなどの遺伝子が含まれる事が明らかとなった。これらはアレルギーの病態においてTh2免疫応答を誘導および活性化する自然免疫機構の重要性を示している。一方，関連領域にはIL18R，IL2RBなどのサイトカイン受容体，活性化した制御性T細胞の特異的マーカーであるLRRC32（GARP）も含まれていた。これらGWASの結果はアレルギー疾患における上皮バリアおよび免疫応答機構の重要性を示唆している。これまで，疫学や免疫研究により，呼吸器感染症，吸入抗原に含まれるプロテアーゼ，大気汚染による上皮傷害は気道アレルギー発症の誘因でもあり増悪要因にもなることが示されている。また，病原体成分や化学物質などのdanger signalsが自然免疫受容体により認識され，Th2免疫応答を活性化させる仕組みも明らかとなってきている。今後もゲノム医学，免疫学，臨床疫学により得られた見識はアレルギーの病態の理解を助け，予防法の確立やより良い治療法の確立に貢献すると思われる。" @default.
