- W23210977 abstract "ABSTRACT Ecdysone, a polyhydroxysteroid insect moulting hormone, which differs in several fundamental aspects from vertebrate steroid hormones, is secreted in large amounts by the ecdysial glands of immature insects. Circulating ecdysone is readily 20-hydroxylated by the Malpighian tubules and the fat body into 20-0H-ecdysone; as a result, ecdysone and 20-0H-ecdysone are both present in the blood and it is as yet unresolved whether the responsive tissues respond to either or both compounds. We have measured ecdysone and 20-0H-ecdysone by radioimmunoassay in the blood and epidermis of last instar larvae of locusts at 6 hour intervals during the 186 hour period comprising this stage. Ecdysteroids appear as three peaks during the fifth instar. The second peak coincides with apolysis and degradation of the inner endocuticle. By prothoracic gland extirpation or X-irradiation of haemopoietic tissue, we have determined critical periods for apolysis and endocuticle degradation. Larvae were deprived of endogenous hormone by extirpation of the ecdysial glands. Binding sites of high affinity and low capacity were identified using radiolabeled ecdysone and 20-0H-ecdysone. We have examined correlations between circulating ecdysteroid levels between wind epithelial ecdysteroids and the appearance of chitobiase activity in the moulting fluid. Fluctuations of chitobiase activity are related to the appearance of ecdysteroid peaks. It appears that 20-0H-ecdysone causes apolysis in vitro in pre-apolytic wing epithelium; other related ecdysteroid derivatives do not." @default.
