- W2322010051 abstract "Event Abstract Back to Event Temporal analysis of multiple co-released neurotransmitters Mária Baranyi1* and Beáta Sperlágh1 1 Hungarian Academy of Sciences, Institute of Experimental Medicine, Hungary Neurons, the cells in the brain communicate chemically with each other by releasing small amounts of chemicals. The composition of the signaling molecules that they release in response to various stimuli is a complex cocktail of neurotransmitters. We developed a sensitive analytical method to investigate how the concentration of signaling molecules changes under the pathologically relevant conditions. The major modifications incorporated into this method include: the reaction of dansyl amide and dansylsulphoarachidonic ester were accelerated by high pH value; modification of the gradient profile to elute amino acids, monoamines and common endocannabinoids within the same run; addition of steps for sample enrichment and cleaning the excess of dansylchloride within each run; shorter re-equilibration time. The quantitative analysis study in rat striatum revealed the interval of extra cellular levels of amino acids, monoamines and endocannabinoids between 0.5 and 250 nM. Upon rotenone treatment, an in vitro model of Parkinson’s disease (PD), the electric field stimulation increased the concentrations of dopamine (DA), glutamate (Glu), serotonin (5-HT) and 2-arachidonylglycerol (2-AG) in the slice perfusate. When, peroxide stimulation was applied in association rotenone treatment with D2 type dopamine receptor agonist, bromocriptine revealed increases of DA, 5-HT and 2-AG, while the Glu, GABA and anandamide (AEA) no alterations in basal levels in slice perfusate. In line with above line of D2 agonist, ropinirole elevated the concentrations of DA, noradrenalin (NA), 5-HT and Glu. In conclusion, the development of this method allows the direct, temporal analysis of co-released neurotransmitters in tissue over flow-fluid and increases our understanding of changes with neurodegenerative diseases.
