- W2326155220 abstract "FIGURE I. The concentration offructosamine measured in dilutions ofhuman serum a/one and with drug added at 0·5 and J·OmM concentrations. NaCl) at O' 5 mM and 1·0 mM concentrations and assayed as before. At low protein concentrations penicillamine produced positive interference, however as the protein concentration increased the interference became negative. The interference was dependent on drug concentration at all protein concentrations. Captopril interference was positive at all protein concentrations and was also dependent on drug concentration but diminished as the protein concentration rose (Fig. I). The drug concentration at which interference became statistically significant is much higher than the therapeutic concentrations (about 4 JiM for Captopril and 70 JiM for penicillamine) of these drugs.P The fact that penicillamine interference is dependent on protein concentration is probably a consequence of the high degree of drug/protein binding (about 90 %) through disulphide bonds. The majority of the remainder forms disulphide bonds with other compounds and less than I % is present in the free form. Positive interference in the fructosamine assay is seen only when there is a possible excess of free drug (i.e. at low protein concentrations), Negative interference may be caused by the site of binding of penicillamine being close to the site of glycation and therefore Additional key phrases: captopril; penicillamine; propylthiouracil" @default.
