- W2327470887 abstract "Purpose: The long-term goal of our research is the development of non-invasive MRI techniques for monitoring muscular dystrophy progression that could be suitable as outcome measures for clinical trials. Here, we evaluated in-phase and out-of-phase and fast imaging with steady precession (FISP) MRI sequences in the estimation of muscle fat content in patients with oculopharyngeal muscular dystrophy (OPMD). Materials and methods: Informed consent was obtained in written from all included patients. Approvement by the local ethical committee was obtained for this prospective study protocol. A total of 9 OPMD patients underwent 1.5-T MRI, and axial T1-weighted, in-phase and out-of-phase and FISP images of the thighs and calves were obtained. Parameters of the FISP sequence were optimized to yield maximal fat-muscle contrast. Fat indexes were calculated from the signal intensity measured on the images. Furthermore, a spin echo (SE) sequence was performed and T2 values were determined using a nonlinear least squares fitting routine. The Pearson correlation coefficient was used to study the relationship between the obtained parameters of MFC and the T2 measurements. Results: Fat indexes from differently affected muscles calculated from in-phase and out-of-phase images correlated with MFC measured using FISP sequences and with T2 measurements. All three indexes were superior to visual estimates using previously published scales. Conclusion: In-phase and out-of-phase, and FISP imaging as well as T2 measurements can all be used to estimate the muscle fat content in patients with muscular dystrophy. Further, imaging at later stages will help to track progression in our patients and tell if quantitative MR imaging could be useful as a non-invasive measure for future therapeutic trials in muscular dystrophies." @default.
