- W2327832236 abstract "1990年に幹細胞増殖因子 (stem cell factor; SCF) が同定されて以来, SCFはヒト肥満細胞に対し唯一の増殖因子として, そして遊走因子および活性因子としての役割を担うことが明らかにされた. 著者らはこのSCFmRNAが鼻粘膜上皮細胞に発現し, この発現が鼻アレルギー患者に強く, また上皮細胞からのSCF産生量の多いことを明らかにし, 鼻アレルギーの発症に重要であることを報告した.今回, 下鼻甲介粘膜広範切除術を行った患者を対象に, (1) 鼻粘膜上皮より単離培養された上皮細胞中の混入細胞の有無 (2) 鼻アレルギーの感作程度と培養上皮細胞からのSCF産生量の相関性 (3) 現在アレルギー治療に使用されている薬剤の培養上皮細胞からのSCF産生に対する抑制効果について検討した. この結果, (1) 単離培養された細胞はサイトケラチン陽性細胞がほとんどであり, 混入細胞は鼻アレルギーにおいて肥満細胞を標識するAA1陽性細胞がわずかに認められたのみであった. (2) 培養上皮細胞からのSCF産生量は, 非アレルギーとダニCAP-RASTクラス1-2群, 3-4群, および5-6群との間に有意差があり, またクラス1-2群と3-4群および5-6群との間に有意差があった. (3) 上皮細胞からのSCF産生に対しフルチカゾンは10-7mol/L以上, プレドニゾロンは10-6mol/L以上, シクロスポリンは10-9mol/L以上でそれぞれ有意に抑制した. クレマスチンは10-7mol/L以上で, ケトチフェンは10-5mol/Lで有意に抑制がみられた. しかしクロモグリケートは10-7～10-4mol/Lで, スプラタストは100ng/ml, 500ng/ml (最終濃度) で共に抑制効果はみられなかった. このSCF量の抑制は細胞増殖を抑制するのではなく, 細胞内におけるSCF産生を抑制することを明らかにした." @default.
