• W2328443270 abstract "Laccase (EC 1.10.3.2)는 구리를 함유하는 multi-copper blue oxidase의 일종으로 고등식물, 곤충, 세균, 균류 등에서 발견되어 왔다. 이 효소는 monophenols, polyphenols, methoxy-substituted phenols, aromatic amines, lignin을 포함하는 많은 페놀 화합물들을 산화시키므로 다양한 기질 특이성을 가진다[9, 13, 26, 30]. 대부분의 laccase는 4개의 구 리이온을 함유하며 단백질 구조 내에 3가지(type-1, type-2, type-3)의 구리결합자리를 가지고 있다. 효소반응에서 기질 의 산화반응은 type-1 구리에 의해 촉매 되어 전자는 trinulear cluster (1개의 type-2 copper와 2개의 type-3 cooper로 구성)을 따라 이동하고, 산소분자의 환원과 물 분 자의 방출을 동반하게 된다. 이런 전자이동 메커니즘을 바탕 으로 laccase는 monomer의 교차연결, polymer의 분해 및 방 향족 화합물의 개열에 관여한다. 이와 같이 laccase는 다양 한 기질 특이성뿐만 아니라 폭넓은 효소 반응성 때문에 염 료 탈색[4], 펄프 표백[21], 생물학적 환경 정화[18], 폴리머 합성[10], 바이오센서[25] 및 바이오 연료전지[16] 등의 여러 분야에서 활용되고 있다. 리그닌은 풍부한 방향족 중합체로 구성되어 산에 의해서 도 쉽게 분해되지 않는 난분해성 물질이지만 백색부후균이 생산하는 laccase, lignin peroxidase, manganese peroxidase 등의 효소들이 리그닌을 분해하는 것으로 알려져 있다. 따라 서 이들 효소를 이용하여 PAHs (polycyclic aromatic hydrocarbons) 또는 chlorophenols 등 여러 환경오염물질의 제거 및 감소에 응용하고자 하는 연구가 진행되고 있다. 특 히 균류의 laccase를 이용한 연구가 가장 활발하게 진행되어 Trametes versicolor 및 Myceliophthora thermophila에 의 한 PAHs의 분해[1], Panus tigrinus 및 Coriolus versicolor 에 의한 chlorophenols의 transformation [15], Trametes villosa에 의한 bisphenol A의 분해[8] 등이 보고되어 있다. 버섯은 자실체로부터 항암, 면역증강, 항균 및 혈압 상승 억제 등의 효능이 밝혀져 식품 및 의약품으로 이용되고 있 Optimal Conditions for Laccase Production from the White-rot Fungus Marasmius scorodonius Su-Jin Lim and Sung-Jong Jeon* Department of Biotechnology & Bioengineering, Department of Smart-Biohealth, Dong-Eui University, Busan 614-714, Republic of Korea" @default.
