Amplification of HER-2/neu oncogene has become an important biomarker for identifying patients who respond to HER-2 targeting therapy. several studies have analyzed HER-2/neu overexpression in gastric carcinoma, and the rate of HER2 positivity is variable, ranging from 6% to 35%. Our goal through this study is to evaluate the HER-2/neu overexpression in our country by immunohistochemistry (IHC) method in all gastric carcinomas samples diagnosed in our department without distinction of staging or histological WHO and Lauren's classification. Methods: It's about an epidemiologic study over 2010 and 2011, HER-2/neu overexpression was examined with semi-quantitative standardized and automated IHC in 78 formalin-fixed paraffin-embedded gastric carcinoma samples by using Ventana Pathway® clone 4B5. Results: Over a period of two years (2010 and 2011), 78 cases of gastric carcinomas were registered in the Department of Pathology, Ibn Rochd Annaba University Hospital - Algeria. Males were more involved than females (2 males/ 1 female), the pathological antecedents of our patients were dominated by the chronic B gastritis caused by HP. The global age average was of 57.64 years with extremes of 24 years and 87 years. Biopsies represent more than two thirds of cases (66.67 %) Gastrectomy specimens represented the third of cases of which half was total gastrectomies. The location of the tumor was not clarified in 42.3 % of the cases which were all cases of biopsies, it was clarified in 57.7 % and showed tumors located in distal stomach in 46.66 % of cases, in corpus in 26.66 %, sub-cardial tumors in 15.55 % of cases and diffused tumors to all stomach in 11.11 %. Histopathological study showed, according to Lauren's classification, that it was about intestinal type in 55.12 % and diffuse type in 42.3 %. Of the 78 gastric carcinoma samples examined by IHC, a total of 07 cases (9%) were positive for HER-2/neu overexpression at a score of 3+. 3 cases were scored 2+ for whom we did not perform CISH or FISH amplification verification, and 5 cases were scored 1+. Conclusion: HER-2/neu overexpression is estimated at 9 % in our department, this rate is included meanwhile world studies. HER-2/neu overexpression may be used as an independent prognostic factor for gastric cancer patients. IHC 2+ cases should be further detected by FISH to assess HER-2/neu gene status. Patients with HER-2/neu amplification also might constitute potential candidates for targeted therapy with trastuzumab especially for the advanced stages." @default.
