- W2334676327 abstract "ISEE-402 Introduction: Asthma is a complex multifactorial disease with a genetic predisposition, immunological aberration and involvement of environmental factors. Polymorphisms of the Glutathione-S-tranferases (GSTs) are known risk factors for some environmentally related diseases. In this study the hypothesis that polymorphisms in the GSTM1 and GSTP1 genes are associated with respiratory outcomes in a susceptible population was examined. Methods: This study adds genetic analysis to the epidemiological study to determine the health status of the South Durban communities exposed to environmental pollutants. The study population consisted of 380 schoolchildren, from the south and north of Durban (a highly industrialised region in the south compared to a similar socio-economic but not industrialised region in the north). The study population consisted of randomly selected grade 4 pupils, together with additional known persistent asthmatics. GSTM1 genotyping performed by multiplex PCR enabled the precise discrimination of individuals homozygous for the null allele. To confirm successful amplification, a set of primers that amplified the b-globin gene was included in each PCR reaction as an internal control. PCR-RFLP analysis with Alw261 was used to determine the genotype at the GSTP1 locus. The reaction successfully differentiated between the Ile/ile, Ile/val and the val/val genotypes. Results: The preliminary analyses of the data suggest a high prevalence of respiratory diseases in the south communities, and the risk factors for this still have to be evaluated. The GSTM1 gene deletion (GSTM1null) was detected in 29.3% of the study population while the GSTP val/val polymorphism was detected in 16% of individuals. Analyses of the role of these polymorphisms in modifying response to environmental exposures will be conducted. Conclusion: Frequencies of the polymorphisms are consistent with frequencies in similar ethnic groups. Analyses of whether GSTP1 and GSTM1 modify air pollution response will be conducted." @default.
