Leuven, Belgium Structural integrity of brain white matter declines in old age, with the most pronounced changes occurring in tracts connecting anterior brain regions. Despite observations that inhibitory control is diminished in the elderly, it is not known whether this is related to the deterioration of frontal white matter, and whether this is specific to task relevant pathways. Right-handed participants (20 old and 20 young, mean age 69 and 25 years) performed a stop-signal task probing both non-selective and more selective aspects of behavioural inhibition. Go trials (70%) required a simultaneous response by both hands. Stop trials required the rapid inhibition of both (Stop All, 10%), or just one (Stop Left, 10%, Stop Right, 10%) hand. As expected stop-signal reaction time (SSRT) was worse in the older group and interference effects were greater during selective stopping. When young adults perform stop-signal tasks, activation of right inferior frontal cortex (IFC), pre-supplementary motor area (preSMA) and subthalamic nucleus (STN) is observed. We therefore outlined STN manually on enhanced contrast anatomical scans, performed probabilistic diffusion tractography, and extracted measures of tract strength and integrity to investigate brain-behaviour relationships. Structural integrity (fractional anisotropy, FA) of white matter associated with right (but not left) IFC, preSMA and bilateral STN regions of interest was related to individual differences in SSRT and interference effects. Bilateral preSMA-STN tract connection strength predicted SSRT and this relationship was strongest in the elderly. This result highlights the importance of bilateral preSMA-STN projections in reactive stopping and links age-related decline in inhibitory control with structural decline of this pathway.
