- W2335895242 abstract "Colorectal cancer (CRC) is one of the most common malignancies.Unfortunately a significant proportion of surgically curedpatients in the early stage of the disease develop progressionand die from the disease. Lymphatic metastasis stronglydetermines treatment algorithms in CRC. Currently,postoperative histology results are needed to determine lymphnode status. Reliable preoperative information would be usefulto advance treatment strategies. Twelve patients (7 withclinical stage II and 5 with clinical stage III with detectablelymph node metastasis) aged 52-76 years, with no neoadjuvantchemotherapy or radiotherapy and histologically confirmed colonadenocarcinoma were included in the study. Relative geneexpression levels of 96 genes encoding extracellular matrix andcell adhesion proteins were obtained by commercial low-densitycDNA microarrays from 12 primary colon cancer samples. Geneexpression data analysis based on the SAM (SignificanceAnalysis of Microarrays) and t-test (alfa = 0,01) methodsidentified 6 genes with significantly different expression inassociation with lymph node status. In subsequent clusteranalysis this group of genes was able to discriminate lymphnode positive from lymph node negative patients. Thrombospondin1, fibronectin 1, osteonectin and TIMP1 genes weredown-regulated and laminin B2 and integrin alfa6 genes wereup-regulated in the group of tumors developing lymph nodemetastasis. All bioptic samples were assessed by pathologistand fraction volumes of particular cell populations weredetermined (malignant cells, lymphocytes, fibroblasts,granulocytes) in percentual values. Interestingly, remarkablefraction volumes of neutrophil granulocytes and related tumorinflammation rates were identified exclusively in the group ofpatients with good prognosis (disease free survival longer than36 month). Extracellular matrix gene expressions were analyzedalso in relation to fraction volumes of particular cellspopulations and significant changes were identified. Ourpreliminary data confirm pivotal influence of others thanmalignant cell populations on gene expression profiles and alsoon tumor phenotype. Analyzing of gene expression data fromlarger group of CRC patients and validation of results by othermore accurate technology (RT-qPCR, imunohistochemistry) willprovide preoperative determination of lymph node status ofcolorectal cancer patients and more individualized therapy inthe future.
