- W2338286390 abstract "Fish semen freezing with liquid nitrogen it’s a valuable technique to preserve genetic material which can be used to genetic improvement programs of economic interest species, besides; it allows the material storage for a long-term period. However, each specie requires a specific protocol. The aim of this study was to develop a semen preservation technique to the Japanese ornamental fish Carassius auratus through the cryopreservation process. The sperm needs the addition of cryoprotectant solutions to its appropriate freezing. As the aim of cryopreservation it’s to obtain viable sperm which keeps its fertility, the motility analysis, postfreezing, it’s one of the most used practices to examine the viability of cryopreserved sperm. So the present study used the motility rate and time to evaluate the effect of semen exposition to different concentrations of the cryoprotectant dimetilsulfoxide, the effect of semen exposition to different activating solution (NaCl 0,3%, NaHCO3 1% and distilled water), and the difference of sperm motility between thawed semen and fresh semen. In order to do it, it was used a optic microscopy linked to a digital camera which registered the sperm activity through video. Statistically, there was no difference in the effect of the different concentrations of DMSO and activating solutions in the range and time of motility of fresh semen, however the 10% DMSO concentration and the NaCl 0,3% activating solution demonstrated a bigger average tendency. Therefore, in the semen freezing experiment, DMSO 10% was used as external cryoprotectant, and NaCl 0,3% as the activating solution. Fresh semen also activated with NaCl was used as control. Both treatments showed difference, whereas control demonstrated a bigger sperm motility range," @default.
