- W2339450767 abstract "We previously suggested that serotonin (5HT) in the mouse stomach is largely of mucosal mast cell origin (7). In this study, we attempted to obtain further evidence for the existence of 5HT and its biosynthetic enzyme, tryptophan 5-monooxygenase (EC 1.14.16.4), in mucosal mast cells of mouse stomach. An athymic nude mouse (BALB/c-nu/nu) and a mast cell-deficient (WBB6F1-W/WV) mouse were used as experimental animals which may have abnormalities in the differentiation or number of mast cells. In the stomach of the nude mouse, the 5HT concentration and the tryptophan 5-monooxygenase activity were 8.8 μg/g wet tissue and 3.0 pmol/min/mg protein, i.e., 1.9-fold and 1.5-fold higher than in the control mice, respectively. In the small intestine and colon, the values were not noticeably different between the mutant and control mice with the exception of a decrease in the 5HT content in the colon of the mutant mouse. The tryptophan 5-monooxygenase activity in the stomach of the mast cell-deficient mouse was 1.2 pmol/min/mg protein, i.e., 46% of that in the control mouse. These results, together with previously published findings on the organ distribution of 5HT in the mast cell-deficient mouse (7), suggested that large portions of 5HT and its biosynthetic activity in the mouse stomach are of mucosal mast cell origin." @default.
