Thus, the drug release rates could be also altered concurrently.Therefore, in this work, we employed an implantable ALZET ® Osmotic Pump, which has been widely used to locallydeliver various therapeutic agents and examined the effect of pH, temperature and ionic concentration on its drugrelease rate. For this, we performed in vitro cell tests to simulate the condition of local tissues influenced by thealtered drug release rates, where we used diclofenac sodium as a model drug. Key words: drug delivery, osmotic pump, pH, temperature, ionic concentration I. 서 론 약물전달시스템(Drug delivery system)은 인간의 질병치료 효과 및 효능을 극대화하기 위하여, 약물을 효율적으로전달해 체내에서 약물의 생체이용도(bioavailability)를높이고, 체내에서의 약물에 의한 부작용을 줄이기 위한 약물의 제형 및 제제를 개발하는 기술이다[1,2]. 특히, 이러한약물전달시스템을 구현하기 위해서 정제, 캡슐, 주사제, 패취제 등이 개발되어 사용되고 있으며, 이와 더불어 이식형약물전달 기기들도 연구 및 개발되고 있는 추세이다[3-5].대표적인 예로, 고분자 기반의 서방형 약물 전달(sustaineddrug release)시스템의 경우에는 약물이 탑재된 고분자 분해(degradation) 또는 약물의 농도 구배에 따른 확산(diffusion)에 의해서 약물이 방출되기 때문에, 한 번의 투여로 장기간 약물을 전달할 수 있다는 장점이 있으나[6,7]정교하게 약물행하였음 방출형상을 제어하기 어렵다는 단점도 가지" @default.
