- W2347052461 abstract "The ready availability of [60]fullerene (C60) and its homologues has increasingly invited exploration of their outstanding new physical and chemical properties. Among them, their easy excitation by visible light irradiation provides great potentials in the application of water-soluble fullerenes for various medicinal uses. For example, they act as a singlet oxygen (1O2) photosensitizer to cleave DNA and as photoinactivator of viruses. In spite of their high potential, however, the applications have been very limited because of their poor water-solubility. Several lines of effort have so far been devoted toward compensation for this drawback, for example, by introduction of water-soluble substituents, mixing with water-soluble polymers or lipid membranes, solubilization in -cyclodextrin ( -CDx), and watersoluble calixarenes. From these methods, we employed the lipid membranes-incorporated C60, which are forming vesicles, by the following three reasons; (i) unmodified C60 in vesicles can generate 1O2 by energy transfer or C60 anion radical by electron transfer more efficiently than other chemically-modified C60 derivatives, (ii) various vesicles with positively charged, negatively charged, nonionic and zwitter ionic surface was prepared by a selection of lipids, for example, phospholipids, aminolipids and glycolipids, etc., and may confer a function as a drug carrier and (iii) the formation of the vesicle with the relatively large size are promising for the enhanced permeability and retain (EPR) effect, which was known that macromolecular compounds or aggregates can preferentially acumulate in solid tumor due to the enhanced permeability of tumor vascular endothelium and and the lack of functional lymphatic drainage in the tumor tissue.
