- W2347158358 abstract "Magnetic particles have gained increasing interest as smart materials because of their potential to be applied in different fields including separation, controlled drug delivery, catalysis carriers etc. [1-2]. Proper surface functionality and colloidal stability of the magnetic particles are key requirements to enable physical or chemical interaction with biological entities such as enzymes, nucleic acids, antibodies for biomedical purposes. Particles exhibiting magnetic and stimuli-responsive properties are promising for controlled switching use. Because of the coil to globule transition upon temperature variation, poly (N-isopropylacrylamide) (PNIPAm) hydrogel coatings on magnetic particles have already been reported. Nonetheless, only few reports have been dedicated to functionalized PNIPAm magnetic particles for nucleic acid (cfr. DNA and RNA) separation. The use of nucleic acids, more specifically DNA/mRNA, as a diagnostic and therapeutic tool has already shown great potential in the biomedical field [3]. The aim of the present work included the development of functionalized, sub-micron magnetic polymer particles that can be used in nucleic acid separation, purification and concentration. Herein, we report on the synthesis, the characterization and the application of functional, pH- and temperature-responsive (Fig. 1a) magnetic polymer particles. Divinylbenzene (DVB) cross-linked magnetic particles were first prepared through emulsion polymerization in the presence of magnetic nanoparticles. Next, PNIPAm shells with different functional monomers have been applied on magnetic/P(DVB) particles using seed precipitation polymerization. The resulting particles were highly temperature-responsive and core-shell structured [4]. In addition, a feasibility study for DNA and RNA capture was performed using the stimuli-responsive magnetic polymer particles.
