- W2347360084 abstract "Objective:To explore the characteristics of ion channel and electrical remodeling with a rapid paced primary cultured atrial myocyte model for atrial fibrillation.Methods: Primary rat atrial myocytes were cultured and a rapid paced primary cultured atrial myocyte model was established.Atrial cells were cultured in the presence of rapid field stimulation(600 beats per min,1.5 V/cm) for 24 h.Action potential durations were recorded from stimulated cells,as well as control cells cultured with the whole-cell voltage-clamp techniques.Expression of L type calcium channel and KV 4.3 were identified by immunocytochemical analysis.Results: After four days of primary culture,the number of cell reached(2～3)×106/ml.The immunocytochemical analysis showed that over 90 percent of cells were stained positively for α-actin,a factor specific for cardiomyocytes.The action potential duration was shortened through the rapid pacing compared with those before pacing(p0.05).The expression of L type calcium channel and KV 4.3 were obviously down-regulation though.Conclusion: Rapid stimulation of cultured atrial cells produces ion channel and electrical remodeling.It is considered that a rapid paced cell model was established with primary cultured atrial myocyte and it can be used to study the mechanism of early electrical remodeling for atrial fibrillation." @default.
