- W2347448892 abstract "Objective To elucidate biological functions of different protein S100A4 discovered from HCC cell lines by comparative proteomics approach.Methods The different expression level of S100A4 among HCC cell lines was validated by RT-PCR, western blot and cell immunochemistry.The expression recombinant plasmid of pcDNA3.1(+) AS S100A4 was constructed successfully and transfected into high metastasic cell lines MHCC97H to observe the alteration of biological functions of MHCC97H.Results The overexpression of S100A4 in high metastasic HCC cell lines was confirmed by validation tests. After transfected with pcDNA3.1(+) AS S100A4, the average amount of invaded cells in cell invasion assay in vitro were 5.0±1.4(MHCC97H/pcDNA3.1(+) AS S100A4),16.4±1.6 [MHCC97H/ pcDNA3.1(+)]and 16.9+1.5(MHCC97H ),while the average amount of invaded cells in cell motility assay were 11.1±3.3 (MHCC97H / pcDNA3.1(+) AS S100A4),18.9±2.0[MHCC97H /pcDNA3.1(+)]and 21.9±3.4(MHCC97H).The quantitative analysis of MMP9 in culture media with serum free by ABC-ELISA displayed 18.2(MHCC97H / pcDNA3.1(+) AS S100A4),28.0 [MHCC97H/ pcDNA3.1(+)]and 31.9 ng/ml(MHCC97H).The concentration of MMP2 were also measured as follows: 29.8(MHCC97H/pcDNA3.1(+) AS S100A4),26.4(MHCC97H/pcDNA3.1(+))and 26.5 ng/ml(MHCC97H). MMPs in culture media with serum free from MHCC97H transfected were detected by Gelatin zymography. The activities of MMP9 secreted from MHCC97H with S100A4 reduction showed lower level than that of controls. Conclusion Some biological properties (including motility, invasion, MMP9 secretion) of MHCC97H transfected with pcDNA3.1(+) AS S100A4 were decreased, whereas distinct changes of MMP2 secretion could not be observed in our experiment. This result implied S100A4 might play an important role in HCC cell invasion and metastasis through two paths of MMPs secretion regulation and motility, invasion potential strengthening." @default.
