- W2347994611 abstract "Objective To investigate the influence of intrathymie inoculation with xenogeneic antigen on the function of recipient lymphocytes,especially natural killer(NK)cells.Methods Three mixed cell cukures were performed using different lymphocytes.The T cells and NK cells from recipients (Rhesus monkey)were,respectively,mixed and cultured with lymphocytes from donors(meishan pig) before intrathymic pretreatment.The same mixed eultures were performed using lymphocytes from recipi- ents after the intrathymic inoculation of donor xenogeneic antigen.After incubation,cells were washed and a 3H-TdR releasing assay was performed to assess the cytotoxicity of NK cells and T cells.The levels of IFN-γand TNF-αin the supernatant were assayed by ELISA.Results Compared with the resuk be- fore inoculation,the secretion of IFN-γand TNF-αof NK cells remained stable after the intrathymic treatment(P＞0.05).But the cytotoxicity of T cells was significantly suppressed(P＜0.05).The levels of IFN-γsecreted by T cells were dropped from(93.9±7.2)ng/L to(79.9±5.8)ng/L(P＜0.05) and the level of TNF-αwas obviously dropped from(19.6±1.9)U/mL to(13.2±2.6)U/mL(P＜0.05).The suppression of T cell function could not reduce the cytotoxicity of NK cells(P＞0.05).Con- clusion Intrathymie inoculation of xenogeneic antigen can specially suppress the cytotoxicity of T cells and decrease the level of eytokines secreted by T cells in recipients,but not influence the cytokine secre- tion of NK cells,suggesting T cells can enhance the cytotoxicity of NK cells through cytokines,but can not decrease the cytotoxicity of NK cells if the function of T cells is suppressed." @default.
