A drastic change in the CD of Chinese cabbage virus (CCV) in saline within therange of pH 3.1~4.8 due to UV irradiation has been observed in our laboratory. Nosimilar phenomenon has ever been reported before. CCV was irradiated with light atwavelengths of 234 and 221nm respectively. The ellipticities of CCV at these two wa-velengths underwent somewhat similar patterns of change: First, the ellipticity roseto the maximum [θ]_(max] with relatively high speed; then it decreased slowly and, aftera relatively long time (120~150 minutes), tended to be stable around a low CD value.Similar phenomena were observed during the irradiation of TMVc or TSV (tomatostreak virus) suggesting its common occurence in the TMV strains. Either in salineoutside the range of pH 3.1~4.8 or in solution with very low saline concentration(e. g. 0.08% NaCl), CCV turns to be unaffected by UV light. Two components ofCCV--the RNA and the coat protein--failed to exibit a similar change in CD whenirradiated under the same conditions. The UV CD spectra of CCV were obtained during the irradiation. The CD spectruminduced by 234 nm light mimiced the shape induced by 221nm light. There wereunusually high ellipticities in the near UV and two strong positive peaks at 230~231nm and 205 nm.The shape of the CD spectra was more or less like those of thearomatic amino acids and some aromatic residue-containing oligopeptides. The possibility that the change in CD observed in this experiment resulted fromthe change in light scattering may be ruled out. It is quite possible that the irradia-tion-induced change in the CD spectrum reflects a change in the structure of CCV." @default.
