- W2348656495 abstract "Objective:To study the effect of free fatty acid on myocardial energy substrate utilization as well as the cardiac function in rat model of type 2 diabetes mellitus. Method:Sprague-dawley rats were conducted into three groups: chow-fed rats were fed normal chow(12% of calories as fat), fat-fed/STZ rats were fed high-fat diet(40% of calories as fat) for 4 weeks and then injected with streptozotocin 35 mg/kg intraperitoneal, fat-fed/STZ/RSG rats were fat-fed/STZ rats treated with Rosiglitazone 3 mg/kg/d for 2 weeks. After measurement of the cardiac function include left ventricular end-diastolic pressure(EDP) and ±dp/dt_ max , the isolated hearts of the three groups were mounted onto a Langendorff perfusion apparatus perfused with KH buffer for 30 min in the presence of 0.4 mmol/L、0.8 mmol/L and 1.2 mmol/L 3H labeling palmitate respectively. Glucose uptake and 3H_2O collection were used to evaluate the rate of carbohydrate and fatty acid oxidation. Result:Compare with the chow-fed group, fat-fed/STZ rats had a significantly depression of glucose uptake of the hearts [(30.7±4.0)∶ (69.0±5.7)μmol/g, P 0.01 ], in fat-fed/STZ group, the oxidative rate of glucose and palmitate was 12% and 88% respectively. Paralleling the reduction was a change of EDP [(14.8±1.9)∶(10.5±1.1)mmHg, P 0.05 ] and-dp/dt [(558±60)∶(650±42)mmHg/s, P 0.01 ]. After 2 weeks treatment of Rosiglitazone, there was a marvelous fallen of blood sugar and FFA in fat-fed/STZ/RSG group compared with the fat-fed/STZ rats. The hearts had an elevated level of glucose uptake ( P 0.01 ), at the same time, EDP decreased from (14.8±1.9) to (11.0±0.8)mmHg/s and-dp/dtmax increased from (558±60 )to (629±51) mmHg/s ( P 0.05 ). Conclusion:Our study indicates that a higher FFA results in a depression of glucose oxidation and a increase of fatty acid oxidation in type 2 diabetic heart. Elevation of insulin sensitivity using rosiglitazone early would increase myocardial glucose metabolism and brings a benefitial effect on heart function." @default.
