- W2350582512 abstract "To identify the cis-acting sites required for the regulation of puc gene expression by O2, seven mutations andfive 10-bp deletions were constructed inthe 5' noncoding region of the PUC operon by in vitro manipulation of cloned pucupstream sequence with a modifiedPolymerase Chain Reaction. (PCR) andoligonucleotide-directed mutagenesis(Fig. 7). Transcriptional fusions of thepuc upstream region containing differentais-mutations or 10-bp deletions wereconstructed by using the reportermolecule encoded by lacZ. The role ofthe target site controlling puc expressionwas investigated by analysing the effects of the mutations and deletions onthe β-galactosidase (β-gal) activity resulting from the fusions when present inwild type R. sphaeroides 241 grownchemoheterotrophically and fermentatively. As shown in Table 3, the β-galactivity of PCF611 with A to G transition mutation at - 12 within a regionof dyad symmetry was about 7. 8 foldthat of the wild type fusion construct inthe presence of O2. The puc upstream sequence of PCFK carrying three munitions at - 197, - 193 and - 190 between positions - 199 to - 185 withinanother dyad symmetry GCAGCN5GCTGC produced a β-gal activity 6. 8times as high as wild type activity. Mutations located at the integration hostfactor (IHF)-binding site in either at- 113 or at - 115 as well as at - 119/- 110 of the puc operon resulted in atwo- to threefold increase in the level ofβ-gal activity relative to wild type under aerobic condition, indicating thatIHF binding is necessary for transcriptional regulation by O2. Binding of IHFis believed to induce a DNA bend whichwas proposed to facilitate interaction between the O2-regulatory repressor andthe two regions of dyad symmetry. Inaddition, deletions M (△- 1 38 / - 1 29 ),F (△-279/-270 ), B (△-369/- 360), and A (△- 507/- 498 ) didnot affect the regulation by O2 repression. All these fusions showed almostthe same level of lacZ gene expressionunder anaerobic condition." @default.
