- W2350612842 abstract "Objective To lay foundation for further research on B cell activating factor to the TNF family (BAFF) by preparing the functional recombinant extracellular regions of 134～285 amino acids of human BAFF (rhsBAFF 134-285 ).Methods The total RNA was extracted from fresh human tonsil tissue and the cDNA encoding human BAFF 134-285 was amplified by RT-PCR. After sequencing, the cDNA was cloned into the prokaryotic expression vector pQE-80L and the recombinant vector was transformed into E. coli DH5α. The rhBAFF 134-285 was expressed by the transformed cells induced by IPTG, and was purifid with Ni 2+ -NTA chromatography. The protein was assayed by Western blot and SDS-PAGE. Then the immune activity of the purified proteins was assayed by MTT. Results 459 bp cDNA was amplified by RT-PCR, which was consistent with the sequence in GenBank. Western blot showed that it was the protein of interest. The purified protein could efficiently activate the Raji cell.Conclusion The rhBAFF 134-285 can be highly expressed by E. coli and has biological activity, which may pave the way for future study." @default.
