- W2351545415 abstract "AIM To study the expression of heat shock protein 90β in human gastric cancer tissues and SGC7901/VCR of MDR type gastric cancer cell line. METHODS Immunohistochemical staining and in situ hybridization were used. 70 gastric cancer tissues samples were histologically classified into 13 well differentiated, 32 moderately differentiated, 18 poorly differentiated gastric cancers and 7 mucous carcinomas. samples of 35 non cancerous tissues around the cancer nest, 17 normal gastric mucosa and 23 cases gastritis were studied as controls. Immunohistochemical staining was performed by the streptavidin biotin complex method. Before staining, the formalin fixed tissue sections were incubated with normal goat serum for 10 min, then the sections were incubated with the primary antibodies overnight. The primary antibodies used were 1∶200 dilution of goat anti HSP90 β. The sections were treated with biotinylated rabbit anti goat IgG and peroxidase conjugated streptavidin (1∶100) for 30 min. They were then reacted with diaminobenzidine tetrahydrochloride. In situ hybridization, the denatured probes were added and hybridization was performed at 37℃ for 48 h. The probes were washed with 2× SSC for 15 min, then with 1× SSC for 15 min, and finally with 0.5× SSC for another 15 min. The digoxigenin labeled oligonucleotide probe was detected using the DIG nucleic acid detection kit. The sections were incubated with chromogenic solution containing nitroblue tetrazolium and 5 bromo 4 chloro 3 indolyl phosphate in a dark box for 3 5 h. RESULTS Heat shock protein 90β was mainly located in the cell plasma and was weakly expressed in non cancerous tissues of gastric mucosa. The expression rates of HSP90β in normal gastric mucosa, gastritis and para cancer tissues were 11.8%, 13.0% and 11.4% respectively and no significant difference was found between them ( P 0.05). The expression of HSP90β increased in gastric cancer. The positive rate of HSP90β in gastric cancer tissues was 30.0%, which was higher than that in non cancerous tissues of gastric mucosa ( P 0.05). The expression rates of HSP90β in well differentiated, moderately differentiated, poorly differentiated gastric cancers and mucous carcinomas were 15.4%, 31.3%, 33.3% and 42.9% respectively. The expression of HSP90β in MDR type gastric cell line SGC701/VCR was higher than that in its parental cell line SGC7901. In situ hybridization showed that the positive signal of HSP90β was mainly located in the cell plasma. CONCLUSION HSP90β expression is higher in gastric cancer tissue than non cancerous tissues of gastric mucosa. HSP90β expression is much increased in poorly differentiated cancer tissues. In MDR type gastric cancer cell line SGC701/VCR is higher than that in its parental cell line SGC7901." @default.
