- W2354048216 abstract "Objective To observe the effect of APP17-mer peptide on learning, memory behaviors, mitochondria transmembrane potentials and apoptosis in hippocampual neurons of Kh DIC mice. Methods The 2.5-month-old Kh DIC mice were randomly divided into the model group and the APP17-mer peptide treated group; KM mice were used as a negative control(age and gender matched). The APP17-mer peptide group was injected with APP17-mer peptide subcutaneously(0.34 μg per mouse), three times per week; while the model group and the control group were injected with normal saline. After 25 weeks,water maze test was used to evaluate the learning and memory ability of these mice. Then mitoehondria transmembrane potentials and the percentage of cell's apoptosis were measured by flow cytometry(FCM)respectively. Results (1)The water maze test showed that the full course swimming time[(98. 06±27. 53, 93.18±30.31, 81.65±28.35)s]and numbers of error reaction(6.11±1.76, 6.71±2.05, 5.23±2.13)at the 3rd,4th,5th day were significantly increased in the model group as compared with the normal control group(76.00±19.69, 69.71±24.12, 52.93± 21.23s)and(4.59±2.09, 3.41±2.29, 2.35±1. 11)respectiveiy,(all P0.05). The results of APP17-mer peptide treated mice were the same as those in normal control mice . (2) The neurons in hippocampus of the model mice had lower mitochondria transmembrane potentials (106.0±12.7)and higher apoptotic rate(7.6±1.5) than did the normal control group (189. 5±15.6, 2.8±0.9) (P0.01). Themitochondria transmembrane potentials (160. 3±18. 6) was higher and apoptotic rate (5.5±1.7)was lower in APP17-mer peptide group than those in the model group(P0.05). Conclusions There are the learning and memory deficits in Kh DIC mice with the decreased mitochondria transmembrane potentials and the increased apoptosis in hippocampual neurons of Kh DIC mice. APP17-mer peptide possesses anti apoptosis characters and could correct the abnormal changes, which may be related to its protective effect on learning and memory." @default.
