- W2354996949 abstract "Objective To observe the effects of long-term propofol sedation on cognitive function and the receptor of advanced glycation end product(RAGE) protein expression in rats,whether there is relationship between changes in cognitive function and expression of RAGE protein,in order to provide a basis for the clinical practice.Methods Twenty-eight adult male Wistar rats were selected and randomly divided into a control group and propofol group,each 14.Pre-experiments were done to find the dose of positive reflex disappearance,intraperitoneal injection of propofol was given by 30 mg/kg,and tail vein indwelling venous catheter was left.Then sedative dose of 36-72 mg/(h·kg) was given to experimental group by continuous infusion of 8 hours per day,5 days in a row,the control group given saline solution.one day after drug withdrawal,Morris water maze was applied to observe rats' behavior for 5 days and escape latency and space exploration time were recorded.One hour after the last water maze test,the rat hippocampus was taken and ELISA was used for the determination of the expression of RAGE protein.Results ①Place navigation:compared to the control group,1-4 days escape latency of propofol group was longer;②test results of space exploration:compared with the control group,the space exploration time of propofol group was shorter and the times of crossing the platform were reduced,differences were statistically significant(F=7.04,22.33,P0.05);③ELISA method for the determination of the RAGE protein results:compared with the control group,RAGE protein expression in hippocampus was increased in propofol group with statistically significant differences(F=5.247,P=0.007).Conclusion Long-term sedation of propofol,may impair learning and memory in rats,cause a reduction in cognitive function in adult rats,which may be associated with their increase in expression of hippocampal RAGE." @default.
