- W2355009881 abstract "Objective To investigate the association between 14-3-3 proteins and neurofibrillary tangles (NFT) in brains of Alzheimer’s disease (AD) patients as elucidating the regulatory influence of 14-3-3 proteins on abnormal hyperphosphorylation of tau in AD. Methods The hippocampus from 4 AD cases clinicopathologically confirmed and 4 age-matched controls without neurological disorders were obtained at autopsy. Expressions of 14-3-3 protein and their associations with hyperphosphorylated tau were explored in brains of the controls and AD patients by immunohistochemical, immunoblotting and co-immunoprecipitation methods. Results 14-3-3 isoforms ζ and γ might strongly stain numerous NFT in AD hippocampus, and isoform β might positively immunostain granular cells and a few NFT in hippocampus of AD brains. The results of immunobloting showed that isoforms ζ and γ up-regulated obviously in AD brains and their value density (4570±92 and 5630±40) were 2—3 times higher than controls (1849±65 and 956±140,P0.01,t test). Each of the 3 above-mentioned isoforms was found bound to the hyperphosphorylated tau and formed a complex in AD brains confirmed by co-immunoprecipitation, and PHF-1 monoclonal antibody precipitated more 14-3-3 protein in AD brains (value density 3859±78) than controls (value density 1540±138,P0.05,t test). No expression of isoform σ was observed in both the control and AD brains. The aberrant immunoreactivity of isoforms β,ζ and γ in AD brains was not found in controls. Conclusions 14-3-3 isoforms β,ζ and γ might participate in the formation of NFT in AD brains, implying that they should be closely associated with the phosphorylation of tau. The exact pathway in regulation of tau phosphorylation by 14-3-3 proteins remains to be elucidated." @default.
