- W2355184002 abstract "After thorough study on the production oflipase from Candida rugosa SIPI 2.230 it wasfound a surfactant Tween-80,and triglyceridessuch as soybean oil,sesame oil,linseed oil andolive oil can obviously increase the productionof lipase.Using the following medium andfermentation conditions,Candida rugosa SIPI2.230 can produce 90~100u/ml of lipase(fishpeptone 0.5%,yeast extract 1.0%,solublestarch 2.0%,(NH_4)_2SO_4 0.1%,MgSO_4 0.1%,KH_2PO_4 0.5%,Tween-80 1.0%,soybean oil0.5%,pH 7.0 before sterilization,cultivationat 26℃ for 24 to 32 h).The enzyme was separated from the cellsby centrifugation(4000 r/min,30 min),andthen purified by batchwise adsorption andelution on DEAE-cellulose,Butyl Tyopearl650G hydrophobic chromatography and SephadexG-100 gel filtration chromatography.The enzymewas purified by 66.8-fold with a total activityrecovery of 8.8%.The purified enzyme washomogeneous in polyacrylamide electrophoresis.The lipase(EC 3.1.1.3,)was stable in thepH range of 6.0—9.0 below 37℃.Its optimumpH and temperature were 6.0 and 45℃,respectively.Its activity was stronglyinhibited by Cu~(2+),Zn~(2+),Co~(2+),Ni~(2+) and Cd~(2+)and was weakly activated by Ca~(2+),Mg~(2+) andMn~(2+).It can hydrolyze sesame oil,soybean oil,peanut oil and rapeseed oil very well.Whenolive oil was used as substrate,its K_m valuewas 8.5×10~(-3) M.Its molecular weight was97,000 dalton estimated by Scphadex G-100 gel filtration chromatography.After the lipase in excess reacted separatelywith olive oil or serum triglycerides,theamount of produced glycerol was determinedby chemical method or enzymatic method,respectively.The results showed a linearrelationship between the amount of glyceroland substrate.This means that the lipase canentirely hydrolyze triglyceride to glycerol andhas no α-or β-ester position specificity forsubstrate.The lipase can be used in enzymaticdetermination of human serum triglyceride,and also for food industry and light industry." @default.
