- W2355442973 abstract "Objective:To establish improved animal models for retinal photic injury and observe the pathological changes from retinal injury in rats subjected to intense light conditions.Methods:A light damaging container was designed. 35 healthy adult SD rats were randomly divided into the control group(CON) and groups of 1 day(D1),3 days(D3),5 days(D5) and 7 days(D7) after light injury. The rats in each group were subjected to intermittent light exposure for 7 days,with a daily schedule of 12 hours of light exposure and 12 hours of darkness. Then they were given 36 hours of darkness. The rats in groups D1,D3,D5 and D7 were given 12 hours of intermittent light exposure with 4178 Lux of illumination for 3 successive days,36 hours in total,then they were raised in a normal environment for 1,3,5 and 7 days,respectively. Afterwards,all the rats in each group were anesthetized with 10% chloral hydrate. Eyes were extracted to make retinal paraffin and ultrathin sections for observation under light and electron microscopy.Results:Normal rats had clear layers of retinal structures. The inner and outer segments were in a neat and orderly arrangement,and the inner and outer nuclear layers were neatly arranged and evenly stained. Morphological changes in receptors appeared in groups D1,D3,D5 and D7,and the receptors gradually deteriorated under progressive light exposure. The changes in group D7 were generally similar to those of group D5. The main changes were as follows:the outer nuclear layer became thinner and sparser. The outer segments of the optic rod photoreceptors were in a disorganized pattern,and the stack structures of membranous disks were disorganized. The mitochondria in the inner segments of the optic rods swelled and vesiculated. The nuclear chromatin in the outer nuclear layer migrated toward the center with an uneven distribution.Conclusion:Long(36 hours) intermittent light exposure of 4178 Lux illumination could lead to retinal photic injury in rats,which successfully imitated the retinal photic injury in rats. Morphological changes in the photoreceptors became evident in the early stages of photic injury. The injury gradually became more severe as time progressed." @default.
