- W2356071058 abstract "Objective To evaluate the method of microarray coupled with microemulsions polymerase chain reaction(PCR)in non-invasive determination of fetal gender in first trimester.Methods Plasma samples were obtained from 138 pregnant women at 4-12 weeks of gestation.Fetal DNA extracted from the plasma of pregnant women.Then six Y chromosome special sequences SRY1,SRY2,SRY3,DYS1,DYS14 and DYZ3 as the marker of fetal DNA were amplified by multiplex-PCR in microemulsions,and fluorescence was labeled at the same times,β-globin gene as an internal control.The labeled target DNA was hybridized and detected by the capturing DNA probes on a microarray slide,signals were scanned by ScanArray 5000 and analyzed.The results were compared to fetal gender at birth.Results The earliest plasma sample which we could detect in this study was collected on the 31st day after pregnancy.The six Y chromosome sequences were detected in 76 of 78 male carrier samples,no Y chromosome sequence was detected in 60 female carrier samples.The sensitivity was 97.4% and the specificity was 100% in determination of fetal gender during the first trimester.Conclusion Microarray coupled with microemulsions PCR has excellent diagnose efficacy in determination of fetal gender.The method can be used to determine fetal sex during the first trimester,and it has potential perspective in X-linked genetic disease screening." @default.
