- W2356519024 abstract "OBJECTIVE To investigate the role of endoplasmic reticulum stress(ERS) in adenosine-induced esophageal cancer(EC109) cell apoptosis.METHODS EC109 cells were cultured with adenosine 2 mmol·L-1 for 24-72 h or EC109 cells were cultured with adenosine 0.5-4 mmol·L-1 for 36 h,and cell survival rates were analyzed by MTT.Cell apoptosis was determined by TUNEL assay.The subcellular locations and expressions of ERS-related proteins,such as glucose regulated protein 78(GRP78),caspase 4,caspase 3,transcription factor CHOP and NF-κB were respectively determined by immunofluorescence test and Western blotting.RESULTSThe survival rates of EC109 cells after EC109 cells cultured with adenosine for 24,36,48 and 72 h gradually decreased to(57.7±15.0)%,(56.5±11.1)%,(43.8±5.7)% and(28.8±4.1)%(r=0.9192,P0.01).After adenosine 0.5,1,2 and 4 mmol·L-1 incubation with EC109 cells for 36 h,the cell survival rates decreased to(83.1±11.2)%,(67.9±6.7)%,(55.3±5.0)% and(45.4±5.4)% in a concentration-dependent manner(r=0.8252,P0.01) and the apoptotic indexes detected by TUNEL increased to(15.5±1.1)%,(28.2±0.8)%,(40.1±2.2)% and(50.6±1.3)% in concentration-dependent manner(r=0.874,P0.05).Immunofluorescence assay showed CHOP,caspase 3 and caspase 4 translocating to nuclei and Western blotting suggested that expressions of GRP78,caspase 3,caspase 4,CHOP and NF-κB increased in a concentration-dependent manner with adenosine 0.5,1,2 and 4 mmol·L-1 treatment(rGRP78=0.9471,rcaspase 4=0.8977,rcaspase 3=0.968,rCHOP=0.9762,and rNF-κB=0.9471,P0.05).CONCLUSION Adenosine can induce EC109 cells apoptosis,and ERS pathway may be involved in this process." @default.
