- W2357006945 abstract "Objective To investigate the effect of mild hypothermia on expression of neuroglobin after hypoxic-ischemia brain damage in neonatal rats and study the cerebral protective mechanism of mild hypothermia.Methods One hundred and ten Wistar rats of 7 days old were randomly divided into 3 groups:sham-operated group (n=10),normal temperature hypoxic-ischemia brain damage group (n=50) and mild hypothermia hypoxic-ischemia brain damage group (n=50).Normal temperature group and mild hypothermia group were divided into 5 subgroups respectively at 1,5,15,30,60 minutes after the establishment of rat model,every subgroup including 10 neonatal rats.The rat model of brain damage was established by ligaturing bilateral common carotid arteries.The rats of rectal temperature (TR)of normal temperature group were controlled at range 36~37℃ and TR of mild hypothermia group were controlled at range 32~33℃.Expression of neuroglobin in the brain tissues was detected by RT-PCR and immunohistochemistry methods.Results RT-PCR analysis showed that the expression of neuroglobin mRNA on 1,5,15,30min(A:1.10±0.05,0.96±0.06,0.79±0.06,0.67±0.08)after ischemia in hypothermia group was significantly increased compared with that of the normal temperature group(A:0.98±0.07,0.81±0.11,0.53±0.08,0.36±0.04)(P0.05).The immunohistochemistry suggested that the integral optical density of neuroglobin on 5,15,30min in hypothermia group,IOD:(23.26±2.83、32.06±2.58、19.39±3.16)×105,was significantly increased compared with that of the normal temperature group,IOD:(19.16±2.16,25.15±4.05,13.78±2.40) ×105,(P0.05).Conclusion The expression of neuroglobin in brain tissue increases rapidly and maintains a longer period of time after hypoxic-ischemia brain injury of newborn rats in hypothermia group,suggesting that it may be one of the cerebral protective mechanism of mild hypothermia." @default.
