- W2357967580 abstract "Objective To study the biological significance of the changes of ER cell types in endocrine therapy for breast cancer.Methods In vitro,after treatment with TAM or MA single and three regimens of TAM MA sequential or combination,the contribution of ER in human breast cancer cell(Bcap 37 cell line) was observed by 17 fluoresceinated estrone (17 FE) cytochemical method and their inhibitory effects were detected by living cells counting.Results The percentage of type A ER(+) cells (the entire cell marked with fluorescent brightness) was elevated markedly from 3.8% to 20.7% in TAM single regimen,and accompanied with a significant fall in type C(cytoplasm with brightness) from 31.2% to 4.0%.It indicated that TAM could brought on transfer of ER from cytoplasm to nucleus.But in MA single regimen,there were not more changes in types of ER.In combination regimens,the percentage of ER(-) cells(type D and type E) was elevated(87.0% in TAM→MA sequential 79.0% and 75.3%;in MA→TAM sequential and TAM single,respectively);the last were TAM+MA and MA(71.3% and 69.3%,respectively).It was 65.0% in control group.Correspondingly,The inhibitory effect of TAM→MA sequential was greatest(79.2%, P 0.01),TAM+MA,the second (66.5%, P 0.01),and MA,the smallest (40.3%).Conclusion ①The transferring of ER from cytoplasm to nucleus after TAM treatment is a very importanct biological phenomenon,and corresponds the two step theory of the ER gene modulation.②The changes of ER types accord with corresponding inhibitory effects after treatment with different endocrine therapy.That is probably attributable to the killing of ER(+) cells." @default.
