- W2359647040 abstract "AIM: To observe the expression of mRNA in peripheral blood mononuclear cells (PBMC) from patients with active pulmonary tuberculosis (PTB) and the effects of Bacillus Calmette Guerin Polysaccharide nucleic acid (BCG PSN) on them. METHODS: PBMC were isolated from 15 PTB patients and 15 healthy donors. The cells were divided into three groups: group A (control group), group B (PPD, culture concentration 10 μg·L -1 ), group C (BCG PSN, culture concentration 0.4mg·L -1 ), and cultured in RPMI 1640 at 37 ℃ under 5 % CO 2 for 48 h. The expression of interferon gamma(IFN γ),tumor necrosis factor alpha(TNF α) and interleukin 10 (IL 10) mRNA in PBMC was detected by RT PCR. RESULTS: The expression of IFN γ mRNA in group A from PTB patients was obviously lower than that in healthy donors ( P 0.01 ). The expression of TNF α and IL 10 mRNA in PBMC from PTB patients was similar to that from healthy donors ( P 0.05). There was no difference of the expression of IFN γ and TNF α mRNA in group B PBMC from both PTB patients and healthy donors ( P 0.05); but the expression of IL 10 mRNA in PBMC from PTB patients was significantly enhanced ( P 0.01). The expression of IFN γ mRNA in group C PBMC was enhanced, but still lower than that in healthy donors ( P 0.05). The expression of TNF α mRNA in PBMC was enhanced, compared with that in healthy donors ( P 0.05). There was no change of the expression of IL 10 mRNA. CONCLUSION: The expression of IFN γ mRNA in PTB patients is reduced. BCG PSN can enhance the expression of the IFN γ mRNA in PBMC, but can not enhance that of IL 10 mRNA." @default.
