- W2360610824 abstract "Objective To investigate the changes of Pulmonary TXB_2/6-keto-PGF_1αratio and ET-1 in endotoxin-in- duced acute lung injury and the effects of meloxicam,so as to explore the protective mechanism of meloxicam.Methods Twenty four Japanese flap - eared white rabbits were randomly assigned to three groups:control group,endotoxin - treated group and meioxicam - treated group.ALI models were replicated with intravascular endotoxin injection (700μg/kg weight),In meloxicam - treated group, meloxieam (2.5 mg/kg) was intravenously injected 10 minutes before the administration of endotoxin.In the process of this experi- ment,blood gas and respiratory frequency were measured at 0,0.5,1,2,4 h respectively.After 4 hours,the rabbits were killed. Pulmonary throraboxane B_2 (TXB_2),6 - keto - prostagiandinF_1α(6-keto-PGF_1α),endothelin-1 (ET-1) were measured by ra- dio-immunoassay.Pathological changes of lung tissues were examined and scored down with light microscrope.Results In endotoxin -treated group,respiratory rates significantly increased and oxygen saturation index(PaO_2/FiO_2) significantly decreased.Plumonary TXB_2/6 - keto - PGF_1αratio and the levels of ET-1 were significantly higher than those in control group(P＜0.01).In endotoxin -treated group,Pulmonary edema,hemorrhage were observed.Pathological score also was significantly higher than that in control group(P＜0.01).In meloxicam-treated group,respiratory rates slightly increased and PaO_2/FiO_2 slightly decreased.TXB_2/6 - keto - PGF_1αratio,the levels of ET-1 and pathological score in lung tissues were significantly lower than those in endotoxin-trea- ted group (P＜0.01).Conclusion Meloxicam can down-regulate TXB_2/6-keto-PGF_1αratio and the levels of ET-1 in lung tis- sue,which may possese protective effects on ALI induced by endotoxin in rabbits." @default.
