- W2361874202 abstract "Objective To study the genetic diversity and the gene expression of F-ATPase subunit gene uncD derived from Streptococcus mutans(S.mutans)clinical isolates,furthermore to investigate its influence on the aciduric ability of S.mutans.Methods Thirty-eight S.mutans isolates were derived from 18 strains dis- playing high acid tolerance and 20 strains displaying low acid tolerance.Gene uncD was amplified with specific primers from S.mutans genomic DNA,and the PCR product was analyzed by RFLP and sequencing.The relative expression quantity of uncD gene against the housekeeping gene rec.A was determined by using semi-quantitative RT-PCR two-step method.A gel documentation system and QUANTITY ONES software were used to assay the da- ta.Results It is testified that two genotypes A and B were revealed by PCR-RFLP with AluⅠdigestion.Fisher exact two-tail test showed that the distributions of A and B genotype strains with varies acidurance ability were dif- ferent(P0.05),and the proportion of A genotype strains from high acidurance group was higher than that from low acidurance one.Some of these amplified uncD genes from different group were sequenced and it was testified that there existed variation of AluⅠsites,which does not located at the function region of uncD gene.Conclusion This study indicated that uncD gene of S.mutans F-ATPase obviously displayed genetic diversity and polymor- phism at mRNA expression level existed.The AluⅠ-RFLP genotypes and the expression levels would be related to different acid tolerance of S.mutans strains,in addition,those strains with high acid tolerance ability would in- crease the expression level of subunit gene uncD." @default.
