- W2362269498 abstract "Objective:To study in vitro the reversing effect of Astragalus Mongholicus Bge(AMB) on immunosuppression of colorectal cancer cells(Colon26),and to analyze their targeting molecules.Methods:Supernatants of re-cultured Colon26 after treated by AMB and the corresponding supernatants of Colon26 treated without AMB as control were collected.The concentrations of immunosuppressors(including TGF-β1,VEGF,IL-4,IL-6,IL-10 and PGE2) in supernatants were measured by quantitative ELISA.The effects of supernatants on five immune functions of murine splenocytes(including NK killing and induced transformation detected by MTT,and expressions of IL-2Rα,CD3e+ζ+ and CD3e-ζ+ detected by FCM) were determined.The relationships between down-regulating effects of AMB on immunosuppressors secreted by Colon26 and the immunosuppressions from Colon26 were ascertained by multi-analysis of correlativity.Results:Immunosuppressors were found in supernatants of Colon26,the concentration of TGF-β1 was highest,and the significant inhibitions of five immune functions of murine splenocytes were showed.After treated by AMB,the concentrations of TGF-β1 and IL-10 in supernatant from first re-cultured tumor cells and its inhibition of four immune functions except for IL-2Rα decreased greatly,but the concentration of IL-6 and PGE2 increased highly.Compared with the supernatant from first re-cultured cells,the concentrations of TGFβ1 in supernatant from second re-cultured cells and its inhibitions of CD3e+ζ+ continued decreasing,the concentrations of IL-6 decreased to the level of control,the inhibitions of IL-2Rα began to decrease,the concentrations of IL-10 increased highly.Positive and negative correlations were exhibited on TGFβ1 and PGE2 respectively,which existed among with inhibition of induced transformation,CD3e+ζ+ and CD3e-ζ+expression,and NK killing,rather than VEGF,IL-4,IL-6 and IL-10.Conclusion:AMB can exert down-regulating effects on tumor immunosuppression from Colon26 through reducing its secretion of TGF-β1 or other immunosuppressors.This should be one of novel anti-tumor mechanisms of AMB." @default.
