- W2362378512 abstract "Objective To investigate the anti-tumor effect of different-concentration arsenic trioxide(As2O3) on transplanted human endometrial cancer cell line(HEC-1-B) in nude mice and on the expression of p-ERK,and further explore the mechanism of the action. Methods A model of subcutaneously transplanted tumor of HEC-1-B was created in nude mice that were randomized to five groups as low-dose-,medium-dose-,high-dose-As2O3 groups(1.0,2.5,and 5.0 mg/kg,respectively),normal-saline(NS) group and DDP group.Intraperitoneal medication was given,according to their body weight,for eight days.The changes of body weight before and after medication were measured.The tumors were collected,tumor-inhibiting rates calculated,liver and renal functions as well as blood routine detected.The changes of the cells were observed electron-microscopically;the cycle and rate of apoptosis detected by flow cytometry;and p-ERK by immunohistochemical technique. Results Different concentrations of both As2O3 and DDP showed different extents of suppressing the growth of tumor cells and enhancing apoptosis,in which,the tumor-inhibiting rate for low-dose-As2O3 group was 34.3%,and the expression of p-ERK was 37.5%,with typical sub-G1 peakemerging in this group.The differences between this group and NS group were significant in terms of apoptosis and tumor weight(F=27.20,33.92;q=9.19,13.34;P0.05).Compared with DDP group,the low-dose-As2O3 group showed no significant hematotoxicity,liver and renal toxicity. Conclusion Low dose of As2O3 can markedly restrain the growth of HEC-1-B cells with slight side effects,which is probably associated with blockage of cell cycle and inhibition of the expression of p-ERK." @default.
