- W2362492130 abstract "Objective To observe the effects of recombinant human interleukin-6(rhIL-6) on glucose metabolism and insulin sensitivity in SW872 adipocytes,and observe the influence of JAK2 inhibitors AG490 and PI3K inhibitors wortmannin on the inhibit effect of rhIL-6 on glucose transportin SW872 fat cells.Methods SW872 preadipocytes were cultured in vitro and differentiated into the matured SW872 adipocytes.rhIL-6 was added into the culture medium of fully differentiated adipocytes for 24 hours.The uptake of 3H-2-deoxy-D-glucose was used to observe glucose transport.The levels of glucose transporter 4(GLUT4) on the membrane of cells was detected by flow cytometry.Results 1.Differntiated SW872 adipocytes incubated for 24 hours with different concentrations of rhIL-6 showed that,under basal states and insulin stimulation,1 μg/L rhIL-6 had no effect on the uptake of glucose.But 5,10,20,50 μg/L rhIL-6 significantly decreased uptake of glucose to 16% vs 20%,28% vs 36%,39% vs 50%,47% vs 57% of control levels,respectively.2.After AG490 pre-treatment,under basal states and in sulin stimulation,the inhibit effect of rhIL-6 on glucose transport decreased 20%,33% respectively.Wortmannin can increased the inhibit effect of rhIL-6 on glucose transport,under basal states and insulin stimulation.3.Differntiated SW872 adipocytes incubated for 24 h with 20 μg/L rhIL-6 showed that rhIL-6 decreases the levels of GLUT4 on the membrane of cells.Conclusions rhIL-6 can inhibit glucose transport through decreasing GLUT4,under basal and insulin stimulated,in SW872 adipocytes in a dose-related manner.rhIL-6 decreases insulin sensitivity in SW872 fat cells." @default.
