- W2362826900 abstract "AIM: To observe the effects of Shexiang mixture(SXM) on the infarct size, serum creatine phosphokinase(CPK) and lactic dehydrogenase (LDH) activities and hemodynamics in a rat model of acute myocardial infarction. METHODS: Male Sprague-Dawley(SD)rats were randomly divided into six groups, namely sham-operated group(sham group), myocardial infarction group(MI group), SXM 3.8, 7.6, 15.2 pill·kg -1 groups, and Shexiang Baoxin Wan (SXBXW) 0.6 pill·kg -1 group. They were daily administered intragastrically with 0.5 % carboxy-methylcellulose-Na(CMC-Na), 0.5 % CMC-Na, three concentrations of SXM suspension (38, 76, 152 pills·100 mL -1 of 0.5 % CMC-Na, respectively), and SXBXW suspension (6 pills·100 mL -1 of 0.5 % CMC-Na) for 3 d, respectively. The volume of giving drugs was 10 mL·kg -1. Thirty minutes after last administration, MI was surgically induced by ligation of the left anterior descending coronary artery (LAD). Infarct size, serum CPK and LDH activities, and hemodynamic parameters were measured 24 h after the ligation of the LAD. RESULTS: The infarct sizes of SXM 7.6, 15.2 pill·kg -1 groups and SXBXW group were (15.2± s 2.4) %, (13.5±2.0) % and (18±3) %, respectively, all of them were significantly smaller than that of MI group ((21.9± 2.0) %, P 0.01)). In MI group, serum CPK and LDH activities were remarkably higher than those in sham group(P0.05, P0.01). The increased serum CPK and LDH activities induced by ligation of the coronary artery were significantly reduced by pretreatment with SXM and SXBXW(P0.05, P 0.01). In MI rats, systolic blood pressure(SBP), diastolic blood pressure(DBP), peak systolic left ventricular pressure(Peak), maximum rate of left ventricular pressure rise(+dp/dt max), maximum rate of left ventricular pressure decline (-dp/dt max), myocardial maximal shortening velocity(V max) were remarkably decreased(P 0.05, P 0.01), but left ventricular end diastolic pressure (LVEDP) was significantly increased than those in sham group(P 0.01). However, reduced +dp/dt max and V max could be increased, and elevated LVEDP could be decreased by pretreatment with SXM 7.6, 15.2 pill·kg -1 and SXBXW, significantly(P 0.05, P 0.01). Furthermore, the fall in SBP and -dp/dt max could be prevented by SXM 7.6, 15.2 pill·kg -1(P 0.05) rather than SXBXW(P 0.05). CONCLUSION: Same as SXBXW, SXM could reduce infarct size, decrease serum CPK and LDH activities, and improve cardiac function in a rat model of acute myocardial infarction." @default.
