- W2363584311 abstract "Objective To explore the role and the related mechanisms of L-arginine on pathological cardiac hypertrophy.Methods Male adult Wistar rats were randomized to three groups, control group(12 rats), ISO group(12 rats) and Larginine group(12 rats). Hypertrophic model of rats was established by isoproterenol(ISO). Pretreated with L-arginine,hypertrophy status of rats were determined by hypertrophy coefficient, collagen content and the expression of ANP mRNA. The activity and level of nitric oxide(NO), nitric oxide synthase(NOS), malonic dialdehyde(MDA) and lactate dehydrogenase(LDH) in serum were observed. Results L-arginine decreased the parameter of ventricular mass, heart weight/body weight [(3.52±0.21) mg/g] and left ventricular weight/body weight [(2.39±0.23) mg/g] in L-arginine group were lower than those in ISO group [heart weight/body weight(3.89 ±0.25) mg/g, left ventricular weight/body weight(2.67±0.26) mg/g], the differences were statistically significant(P 0.05). Meanwhile, the relative ratio of the ANP mRNA expression in L-arginine group(1.2) was lower than that in ISO group(1.5), the difference was statistically significant(P 0.05); the myocardial interstitial collagen content of L-arginine group [(14.52±3.09)%] was lower that of than ISO group [(35.24±4.78)%], the difference was statistically significant(P 0.01); the NO content [(34.15±6.12) μmol/L] and NOS activity [(23.9±3.12) U/mL] of L-arginine increased than those of ISO group [(NO content(20.96 ±5.06) μmol/L,NOS activity(16.58±3.12)U/mL], the differences were statistically significant(P 0.01), at the same time, the MDA content [(308.73±37.48) nmol/L] and LDH level [(2065.35±347.46) U/L] of L-arginine group were decreased than those of ISO group [MDA content(389.63±42.85) nmol/L, LDH level(3582.89±364.51) U/L], the differences were statistically significant(P 0.05 or P 0.01). Conclusion Exogenous L-arginine can inhibit cardiac hypertrophy through increasing NO and NOS level, decreasing the level of MDA and LDH, and decreasing myocardial interstitial collagen content." @default.
