- W2363669752 abstract "The experiments which form the basis of this paper indicate that:(1) Formalin and potassium bichromate prove to be the best preservatives of mitochondria even without the admixture of other chemicals. The mixture of the two reagents preserves mitochondria. Even though they are mixed with formalin, potassium bichromate or HgCl_2, they do not change its fixing properties.(2) Saturated solution of HgCl_2 preserves the fixation of mitochondria. When HgCl_2 is used alone, it makes the tissue swell; but being treated with the mixture of formalin with such a reagent, the cells of the tissue retain their original size, as formalin has the function of hardening the tissue. So formalinsublimate is the best fixative for demonstrating mitochondria. At the same time as HgCl_2 precipitates nucleoprotien, it preserves the nuclear structure very well,(3) Fix tissues with formalin of various hydrogen-ion concentrations from pH 1.1 to pH 12.2. The fixation of mitochondria is assured from pH 2.4 to pH 9.0. But on the acid side of pH 2.4 or on the basic side of pH 9.0, mitochondria are destroyed.(4) Fix tissues with saturated solution of potassium bichromate of various hydrogen-ion concentrations from pH 1.1 to pH 7.0. Mitochondria are preserved well from pH 2.4 to pH 6.0. The result of this experiment apparently contradicts Zirkle's opinion. Zirkle considers that the fixation image of potassium bichromate on the acid side of pH 4.5 akin to chromic acid will dissolve mitochondria. But my experiment shows that potassium bichromate at the acid solution will dissociate more dichromate ions than in the basic solution, and so mitochondria may be preserved." @default.
