- W2363841004 abstract "Objective To explore the hypothesis that human cord blood mononuclear cells(HCMNCs) could enter the brain of vascular dementia(VD) rats across the blood-brain barrier(BBB) through intracarotid infusion by observing the characteristics of localization and survival of HCMNCs in the sites of damaged brain.Methods The rats were randomly divided into a control group(n=6),a VD model group(n=6) and a treatment group(n=30).The VD rat model was established by modified pulsinellis 4VO.The rats in the treatment group were divided into 6 h(6-h),24 h(24-h),3 d(3-d),7 d(7-d) and 2 weeks(2-w) group after 4-Vessel occlusion(4VO) with 6 rats in every group and received intracarotid infusion of 3×10~6/0.5 ml HCMNCs labeled with BrdU in vitro.The learning-memory abilities were measured by using computerized shuttle-training case.At 4 weeks after HCMNCs infusion,the characteristics of HCMNCs survival and distribution in brain were studied by immunohistochemical method.Results HCMNCs distributed throughout the brain,and mainly localized in hippocampus and cerebral cortex,especially around their blood vessel.HCMNCs increased from(3 598±311) in the 4VO-6-h group to(4 311±311) in 4VO-24-h group and(4 426±399) in 4VO-3-d group,then decreased gradually to(3 726±356) in 4VO-7-d group and(2 525±298) in 4VO-2-w group(P0.05,P0.01).At 2 and 4 weeks after infusion,the ratio of active avoidance reaction(AAR) was higher in 4VO6-h group than the model group(P0.01),lower than that of 4VO-24-h and 4VO-3-d groups,then decreased in 4VO-7-d group that was significantly lower than that of 4VO-3-d group(P0.05).AAR ratio of 4VO-2-w group continued to decrease, but still higher than than the model group(P0.05,P0.01).Conclusion HCMNCs by intracarotid infusion could enter the cerebra parenchyma across BBB,localized and directed to the damaged sites of brain to survive,and improve learning-memory abilities of VD rats.There is special and suitable therapeutic time window when HCMNCs pass BBB." @default.
