- W2364180589 abstract "ObjectiveTo investigate whether grafting neural stem cells (NSCs) can ameliorate sensorimotor deficits following neonatal hypoxic-ischemic brain damage (HIBD) in rats. MethodsThirty-five 7-day-old SD rats were randomly divided into 3 groups:sham group ( n =10),HIBD group ( n =12),and transplant group ( n =13). HIBD group and transplant group were anesthetized and subjected to HIBD by combination of left carotid ligation and exposure to 8% oxygen for 2 hrs. Media or neural stem cells (NSCs) prelabeling with BrdU were transplanted into the sensorimotor cortex in the hemisphere contralateral to the lesion 3 days after HIBD.Behavior deficits were assessed at 2 to 4 weeks after transplantation. At the end of the behavioral tests, the animals were killed and evaluated for NSC survival and differentiation. Resultstransplant group showed significantly improved sensorimotor function as compared with HIBD group. In T-maze acquisition tests, transplant group had a increased rate of spontaneous alternation[(69.23±13.34)% vs (45.00±27.27)%] ( P 0.05);in the grip-traction test, the time to falling was longer in the transplant group than in HIBD group[(49.15±13.39)s vs ( 27.20± 15.18 )s]( P 0.05). Asymmetries of limb-placing and foot-faults ( P 0.05) were not detected in transplant group, and the motor function was improved in the postural reflex test ( P 0.05). Histological analyses revealed that transplanted NSCs expressed markers for neurons (NF-200). ConclusionThe transplantation of NSCs may improve sensorimotor dysfunction after HIBD.This approach warrants continued investigation in light of potential therapeutic value." @default.
