• W2364221422 abstract "【Objective】 To observe changes in proportion of Foxp3+ CD4+ regulatory T cells(Treg cells)、CD4+CD25+ double positive T cells and T cell subset in peripheral blood of Acute and chronic greenhouse farmer's lung(GFLD) patients and healthy farmers working in plastic film greenhouses.To investigate the effect of Foxp3+ CD4+ Treg cell in the pathogenesis of GFLD.【Methods】 ① By stratified-cluster random sampling,a questionnaire survey analysis for farmers working in plastic greenhouse was performed in Shenyang City,Jinzhou City and Chaoyang City,from August 2006 to June 2009.There was 5880 persons in the survey,and 5 420 effective persons.Response rate is 92.2%.There were 167 fungus greenhouse,126 poultry,208 flowers greenhouse,334 vegetable greenhouse,a total of 835 plastic film greenhouse.There were 231 vegetable greenhouse in Shenyang City and 855 vegetable greenhouse farmers.Screening out 55 cases of patients with GFLD,Rule out 13 cases who do not meet the standards,and eventually enrolled in the study of GFLD patients were 42 cases.The lung HRCT was performed for these patients.Random select 20 cases who have acute GFLD as acute form,22 cases who have chronic GFLD as chronic form,and another 17 healthy farmers as the control group.② Take the median cubital vein blood 2 ml from each subjects in each group at 8:00 in the morning.The proportion of Foxp3+ CD4+ regulatory T cells(Treg cells) and T cell subset in peripheral blood were assayed by flow cytometry method.Comparisons of the data between different groups were performed using one-way ANOVA,and SNK and Games-Howell test was used for comparison between 2 groups.Spearman correlation test was used to observe the correlation between the proportion of Treg cells and T cell subset.【Results】 ① The cell ratio(0.43±0.17)% and(1.10±0.26)% of peripheral blood Foxp3+ CD4+ Treg of the acute and chronic phase group are obviously less than(1.79±0.60)% of matched group,this notable difference has statistic significance(P 0.01).② The percentage of CD4 and CD25 double positive T cells in the peripheral blood of the acute and chronic phase group [(2.70±1.04)%、(2.98±1.35)%] was significantly decreased as compared to control group [(4.72±1.65)%],P 0.01;Compare with double positive cell ratio of peripheral blood CD4 and CD25 of the acute and chronic phase group,the ratio difference has no statistic significance(P 0.05);③ The CD4+CD3+% ratio(19.79±10.96)% and(30.96±10.76)% of peripheral blood of the acute and chronic phase group are obviously decreased than(36.86±11.57)% of control group,especially for the acute phase group.This notable difference has statistic significance(P 0.01).The CD8+CD3+% ratio(36.65±18.71)% and(21.87±7.53)% of peripheral blood of the acute and chronic phase group are obviously increased than(20.15±6.14)% of control group,especially for the acute phase group.This notable difference has statistic significance(P 0.01).Compare with control group,the peripheral blood ratio(0.56±0.18) and(1.43±0.21) of CD4+/CD8+ of the acute and chronic phase group are obviously different,which has statistic significance(P 0.05);④Correlation Analysis: The ratio of CD4+/CD8+ in peripheral blood of the acute and chronic phase group have obvious positive relativity with CD4 and CD25 double positive cell ratio in peripheral blood of the acute and chronic phase group(r value are 0.827 and 0.420,P 0.05);There's no relativity between peripheral blood cell Foxp3+ CD4+ Treg ratio and CD4+/CD8+ ratio and age in the acute and chronic phase group(r value are 0.079 and 0.131,P 0.05).【Conclusion】 The down regulation or overconsumption of Foxp3+ CD4+ Treg cell may be one of key factors of the pathogenesis of GFLD." @default.
