- W2364238596 abstract "AIM To study the pharmacokinetics of acetaminophen (Ace) and the effect of phenobarbital (Phe) pretreatment on the disposition of Ace in mice. METHODS Normal mice and those administered ip Phe 50 mg·kg -1 for 7d,were given iv Ace 100 mg·kg -1 ,and the concentrations of plasma Ace, and urinary Ace, glucuronide Ace(GA) and sulfate Ace (SA) were measured by HPLC method. The concentration time curve of plasma Ace was automatically mimiced with the computer to calculate the pharmacokinetic parameters of Ace, and the differences of the parameters and Ace metabolites between mice pretreated with and without Phe were checked by Students t test. RESULTS The best fits fot the plasma concentration time data of Ace were achieved by the one compartment model, T 1/2 was(13 9±0 55) min and CL(0 0403±0 0038)L·kg -1 ·min -1 . The total recovery of Ace as urinary Ace, GA and SA was 54 0%±6 21% in the urine collected for 9 h. After the pretreatment with Phe, Ace T 12 was shortened by 27 7%, CL increased by 25 2%, and the content of urinary GA decreased by 19 3% (P 0 05). CONCLUSION At least in ICR and Kunming mice, except the conjunction with glucuronide and sulfate, Ace had other metabolic pathway that is very important for the disposition of plasma Ace. The pretreatment with Phe accelerated the metabolism of plasma Ace in mice,but the effect was unrelated to the conjunction with glucuronide and sulfate." @default.
