- W2364276463 abstract "The presumptive ectoderm of thenewt early gastrula is pluripotent.Under different induction conditions,it will differentiate into different struc-tures.We use the method of indirectimmunoperoxidase staining with anti-5-methylcytosine antibody to study thechanges of chromosomal DNA methy-lation during the different developmental stages,so as to understand therelationship between DNA methylationand embryonic differentiation. Eggs of Cynops orientalis served asour experimental material.Threekinds of tissues were used for metaphasechromosome spreadings:(1) the isolat-ed presumptive ectoderm of early gas-trula,(2) the ectoderm of the samestage treated with extract of guinea pigbone-marrow (3 mg/ml) and (3) theisolated neural plate. The results of the indirect immunoperoxidase staining of these chromosomes with anti-5-methylcytosine areshown in figures 1,2 and 3 respectively.Antisera specific to 5-methylcytosine reacts with deoxy-5-methylcytidylic acidresidues in the single stranded DNA inthe chromosome.We have found thestainings of anti-5-methylcytosine arevery light and most of the stainings wereobserved around the surface of the chro-mosome.It seems that very little amo-unt of 5-methylcytosine were presentin all of these three kinds of tissues. If both the control presumptiveectoderm and the ectoderm treatedwith extract of guinea pig bone marrow(3 mg/ml) were continued to be cultured for another 4 days and the isolatedneural plates for 3 days in Holtfretersolution at 20—21℃,when the dif-ferentiations of all these tissues correspond to those of the tail bud stage.The indirect immunoperoxidase stain-ings of anti-5-methylcytosine are muchdarker.Intense staining in telomeresand periphery of the centromers wereusually observed (Figures 4—6).Theamounts of 5-methylcytosine in thesechromosomes are obviously increased.The staining patterns are quite similarto those of adult cells,such as intestinalepithelium,of Cynops orientalis,but itis quite different from the patterns ofthe tissues of early developmental stages. The reasones of differences ofanti-5-methylcytosine indirect immuno-peroxidase staining and the relation-ship between DNA methylation andembryonic differentiation are also dis-cussed ." @default.
